I wouldnt say they are like having flyknits on your feet or anything but they arent UNcomfortable by any means. I didnt have any problem wearing them for an extended period of time. fwiw here is the classic bike in calf skin in a 48 to compare to the plonge lambskin 48 I thought fit was pretty similar. The plonge seemed to sit better on the shoulder.
